In this study, we identified psiTPTE22-HERV as a novel tumor suppressor that was down-regulated by promoter methylation in gastric cancer. psiTPTE22-HERV exerts tumor-suppressive functions by down-regulating EIF4E via inhibition of the PI3K/AKT/mTOR pathway. Furthermore, down-regulation of psiTPTE22-HERV in primary tumors serves as an independent risk factor for poor prognosis in patients with gastric cancer.
